I will be flying from Chattanooga to Ft Lauderdale via Charlotte the first week of January on AA. Will 54 minutes be enough of a layover for my checked baggage to make the connecting flight?

Charlotte only has one Terminal. E and C are concourses. It should take 10-15 minutes at the absolute most.

You will have to go through US Immigration and Customs in Cancun. I recommend at least 75 minutes.

Yes, that is enough time. There is no US Immigration or Customs when leaving the US. All you have to do on your way to Rome is go from your arrival gate to your departure gate.
