First time arriving in Atlanta on international flight (Delta). We have 2hrs 10 min to connect our next flight to RDU. Will this be enough time and what is the customs procedure in Atlanta? Will we have to claim and re-check our luggage? Is arriving on a Tuesday any better than arriving on a weekend?
I am flying in from Manchester with Virgin Atlantic and have a connecting flight to Cleveland Ohio via Delta , will I have to claim my luggage and go through security checks in Atlanta , or will they send my luggage straight on to Cleveland , and I go through security there ?

Yes, in Atlanta, you will go through US Immigration, claim your bags, go through US Customs, re-check your bags at the re-check desk, go through security and take the Plane Train to your departure concourse and gate.

You won't know your gates until the day of your flight. Delta uses every concourse in Atlanta. All you have to do is go from your arrival gate to your departure gate. The Plane Train connects all concourses inside security. You have enough time if the first flight is on-time.
Our flight is from Atlanta to Barcelona but 1 stop in Philadelphia. Will I check in at the domestic terminal since initially flying to Philly or at the International terminal since destination is Barcelona? Thank you!

Yes, you go to the domestic terminal. American only has a check-in desk in the Domestic terminal, so you wouldn't even be able to check in at the International Terminal.
Looking to book a flight on delta from MSP to SXM. The layover in ATL is only 50 minutes. Is that enough time to make the connection to SXM? Will everything have to be on time to be able to make it or is their a bit of flexibility in case we are taxiing, etc when I land from MSP?

Yes, that is enough time. All you have to do is go from your arrival gate to your departure gate using the Plane Train. Arrival times include time spent taxiing (the time shown is the time that it's anticipated you will arrive at the gate, not the time you land).
I'm flying with Delta from Mexico City to Brussels with a connecting flight in Atlanta. I have 1h and 50min. for my connection. Seeing as I will be arriving from MEX, I will need to go through customs. Do I also need to claim my baggage at ATL airport and check in again in the departure hall? Or will my bagage be automatically send to my final destination?
You won't have to claim your bags. Just Immigration, customs, and security. You should have enough time if everything goes smoothly.
